Clinical-stage oncology company Prescient Therapeutics (ASX: PTX) has reached a key operational milestone in its ongoing Phase 2a study of lead drug candidate PTX-100, having met the pre-specified enrolment threshold required for its first planned Dose Optimisation Committee (DOC) review.

The announcement confirms that 20 evaluable patients have now been enrolled in the study evaluating PTX-100 for relapsed or refractory Cutaneous T-Cell Lymphoma (CTCL).

Dose Optimisation Milestone

The Phase 2a study design evaluates two distinct dosing arms, with 10 evaluable patients enrolled in each.

Under the trial protocol, patients are deemed evaluable upon the completion of four treatment cycles.

Reaching this threshold formalises the timing for the study's first planned DOC review, which the company stated remains on track to convene in December 2026, consistent with its target timeline.

Comprising clinical and biostatistical experts, the committee is tasked with reviewing safety and efficacy data from both dosing cohorts to provide guidance on the continuation and design of further study development.

Prescient noted that it will provide a market update following the committee’s recommendations.

The company highlighted that the December review represents a scheduled dose optimisation milestone within the ongoing trial rather than the completion of the Phase 2a study itself.

International patient enrolment continues toward a full target of 40 evaluable patients.

Prescient Chief Executive Officer James McDonnell stated that reaching the enrolment milestone reflects solid execution by the company's clinical team and trial sites as the drug advances toward its next phase.

Compound and Platform Background

PTX-100 is a targeted small-molecule compound designed to block geranylgeranyl transferase-1 (GGTase-1).

By inhibiting GGTase-1, the drug disrupts oncogenic RAS pathways by preventing the activation of Rho, Rac, and Ral circuits in cancer cells, promoting apoptosis (programmed cell death).

According to company filings, PTX-100 is currently understood to be the only GGTase-1 inhibitor undergoing clinical trials globally.

The candidate previously demonstrated safety and clinical activity in Phase 1 trials and a PK/PD basket study, as well as positive efficacy and safety signals in a Phase 1b expansion cohort in T-cell lymphomas.

The US Food and Drug Administration (FDA) has granted PTX-100:

  • Orphan Drug Designation: Covering all T-cell lymphomas.
  • Fast Track Designation: For the treatment of adult patients with relapsed or refractory mycosis fungoides, the most prevalent CTCL subtype.

Commercial Significance

For Prescient, the December DOC review carries meaningful commercial weight as a value-inflection point that de-risks PTX-100's path to market.

A favourable readout on safety and efficacy across the two dosing arms would strengthen the case for advancing into pivotal trials and sharpen the drug's positioning in the CTCL treatment landscape, where relapsed or refractory patients represent an underserved market with limited options.

The existing FDA Orphan Drug and Fast Track Designations already confer tangible commercial advantages—including potential market exclusivity, development incentives, and an expedited regulatory pathway—that could accelerate time to market and enhance the asset's appeal to potential licensing or partnering.
